A. Temperature affects the activity of the enzyme because enzymes are substrate specific so an enzyme structure is made so that it can only work for one substrate (it has a specific function). If an enzyme heats up they start to move faster and collide so its activity increases, but if it gets too hot the enzymes will start to denature because it agitates the polypeptide chain enough to overpower the weak interactions that stabilize conformation and it will lose its structure and therefore will not be able to perform its function. It will not be able to be active and catalyze chemical reactions anymore. The pH affects the activity of the enzymes in a similar way as temperature because it can also help until a certain amount then it starts to denature an enzyme because most enzymes have optimal pH levels. pH levels can disrupt hydrogen bonds, ionic bonds, and disulfide bridges that maintain a protein’s structure and enzymes are proteins.
B. First get a substrate solution and add the specific enzyme that catalyzes the substrate reaction. Have about 5 containers of this solution so that it is possible to test them at different temperatures about 15 degrees Celsius apart. Analyze each container for about a half hour and record the enzyme activity. If the temperature is constantly increased, then the enzyme activity will go up from zero and back down to zero. The independent variable will be the temperature the solution is at, the dependent variable will be the rate of chemical reactions that take place, and the control will be a solution that has not been heated or cooled down but at room temperature. The dependent variable will be measured by calculating how many extra enzymes are left.
